An immunoistochemical (IHC) test was developed to detect bovine respiratory syncytial virus (BRSV) in cell cultures and tissues of experimentally infected mice and calves, using a commercial monoclonal antibody (Mab) against human respiratory syncytial virus (HRSV), as a less expensive alternative, instead of producing specific monoclonal antibodies to BRSV. Clinical samples from calves suffering respiratory disease were also submitted to this test. IHC detected BRSV antigens in mouse tracheas (3, 5 and 7 days post-infection) and lungs (5 and 7 days post-infection), and in one of three lungs from experimentally infected calves. Lungs samples from two naturally infected calves were tested and resulted positive for BRSV by the IHC test. These results suggest... |

Sildenafil citrate is a type-5 phosphodiesterase inhibitor (PDE-5), able to inhibit type-6 phosphodiesterase (PDE-6) as well, providing clinical benefits and paraeffects, some of them potentially related to the retina. The effects of the sildenafil on the retrobulbar and retinal circulation were studied in 27 adult male rabbits of the White New Zealand breed. The electric activity of the retina was evaluated before and at the end of the treatments, and immunohistochemistry studies were conducted. An amplitude increase of the b wave was found in the mixed response of cones and rods after 7 days of treatment with sildenafil citrate. However, in the other evaluations and periods, the values did not differ from the basal ones. Through immunohistochemistry, no... |

Spermatogenesis is associated with considerable fluid secretion or absorption in the male reproductive tract. Aquaporins (AQPs) are membrane protein channels that allow the rapid movement of water through epithelium. In the present study, immunohistochemistry was utilized to localize the expression of AQP 1, AQP2 in the testis and prostate of adult bactrian camel (Camelus bactrianus). Results show that AQP1 have intense reaction in rete testis, efferent ducts, vessels, seminiferous duct and in the prostate, AQP2 was found minor expression in the rete testis, vessels and prostate, which suggesting that AQP1 may have the main role in the absorption of the large amount of testicular fluid in male camel reproductive tract. Investigations of AQPs biology in... |

The extracellular matrix (ECM) plays an important role in the regulation of biological events such as the development of cell migration, proliferation and differentiation. Chronic sun exposure causes changes present in the ECM of actinic cheilitis (AC), a premalignant lesion of the lower lip which helps to understand the carcinogenesis of the lip. This study aimed to investigate elastin, the main component of solar elastosis alternating current in an attempt to establish the relationship between this protein and ECM in epithelial dysplasia. Paraffin embedded tissue sections of the lesions of 35 cases of AC were analyzed by immunohistochemistry for elastin, and became the association with the degree of epithelial dysplasia and age. Highest scores of elastin... |

ABSTRACT: A carcinoid is a rare neoplasm derived from neuroendocrine cells that may arise in the hepatobiliary system. This report described two cases of carcinoid of the gallbladder in dogs, with emphasis on the clinical, pathological and immunohistochemical aspects. Case 1 was a 10-year-old female Shih Tzu, with a history of prostration and occasional episodes of diarrhea and vomiting. Case 2 was a 10-year-old male Cocker Spaniel, with a history of diarrhea and abdominal pain. Abdominal ultrasonography suggested a neoplasm of the gallbladder in both cases. The dogs underwent cholecystectomy, and 10% formalin-fixed samples were sent for pathological examination. Histologically, a neoplastic proliferation of neuroendocrine cells was observed in the... |

ABSTRACT: This report aimed to describe the clinical, epidemiological and pathological aspects of an outbreak of rabies in buffaloes in Rio Grande do Sul (RS), Brazil. Seven out of 41 buffaloes from the same farm became fatally ill; they showed the clinical signs of ataxia, ascending paresis, and paralysis of the limbs. During necropsy, marked hyperemia of the leptomeningeal vessels, and severe distention of the urinary bladder were noted. Histologically, there were indications of nonsuppurative meningoencephalitis and ganglioneuritis with intracytoplasmic and eosinophilic inclusion bodies in the neurons. The fluorescent antibody test was positive for rabies. Viral antigens were also detected by immunohistochemistry (IHC) in sections of the cerebellum.... |

Flow cytometric analysis of DNA content was performed on 28 samples of canine mammary tumors. Nine of them were benign and 19 were malignant. All benign tumors and 11 malignant tumors (57.9%) were diploid (P<0.05). Form the aneuploid tumors, five (26.3%) were hyperdiploid, one (5.3%) hypodiploid, one (5.3%) near triploid and one (5.3%) multiploid. The analysis of the expression of the markers PR and CD31 revealed a significant difference between diploid and aneuploid tumors (P<0.05). The immunoreactivity of PR was higher in diploid tumors, while the immunoreactivity of CD31 was stronger in aneuploid tumors. No difference between the markers MIB-1, c-erbB2, p53 and Cyclin D1 was observed (P>0.05). Using the flow cytometry analysis and... |

The authors describe their experience with an automated immunohistochemical system applied to canine tissue samples. Twenty human cellular markers specific monoclonal and polyclonal antibodies and two different antigen retrieval methods were used in normal and neoplastic breast tissue, as well as skin samples obtained from female dogs of pure and mixed breeds. The antibodies tested were the most frequently used in human and veterinary medicine studies, employed with diagnostic purposes in breast pathology, as well as in cancer research. Most of them may be used to study other normal and abnormal tissues and included cytokeratins, progesterone receptor, c-erbB2, p53, MIB-1, PCNA, EMA, vimentin, desmin, alpha-actin, S-100, pan-cadherin, and E-cadherin. Fourteen vomeronasal organs (VNOs) of adult males one-humped camels (Camelus dromedarius) and Egyptian water buffaloes (Bubalus bubalis) (n=7/each) were examined immunohistochemically with neuronal markers; synaptophysin (SYP) and glial fibrillary acidic protein (GFAP) to clarify the distribution of the vomeronasal (VN) receptor cells and nerve fibers, in addition to elucidate the existence of non-neuronal elements via S-100 and endothelial nitric oxide synthase (eNOS). In both animals, the VNO was lined medially with VN sensory (olfactory) epithelium and non-sensory (respiratory) epithelium laterally. Immunohistochemically, both animals showed SYP immunolabeling only in the receptor cells of VN sensory epithelium while GFAP labeled the ensheathing cells... |
